Theriva Biologics, Inc.
Theriva Biologics, Inc. is a clinical-stage immuno-oncology company dedicated to developing oncolytic viruses designed to overcome the protective barriers of solid tumors and selectively eliminate cancer cells. The company's mission is to improve current standards of care and transform the cancer therapy landscape by enhancing the efficacy of existing treatments. Headquartered in Rockville, Maryland, with a European subsidiary in Spain, Theriva Biologics focuses on addressing areas of high unmet medical need in oncology.
The company's lead clinical-stage candidate is VCN-01 (zabilugene almadenorepvec), an oncolytic adenovirus engineered to replicate aggressively within tumor cells and degrade the tumor stroma barrier, which can impede cancer therapies. VCN-01 is currently under evaluation in clinical trials for various cancers, including metastatic pancreatic ductal adenocarcinoma, retinoblastoma, head and neck squamous cell carcinoma (HNSCC), ovarian cancer, and brain tumors. Theriva Biologics also has other therapeutic candidates, such as SYN-004 (ribaxamase), aimed at preventing microbiome damage and reducing acute graft-versus-host disease (aGVHD) in transplant recipients, and SYN-020, a recombinant oral intestinal alkaline phosphatase, which was recently licensed to Rasayana Therapeutics for gastrointestinal and systemic diseases.
Led by CEO and CFO Steven Shallcross, Theriva Biologics has recently achieved significant milestones, including a positive End-of-Phase 2 meeting with the U.S. FDA for VCN-01 in metastatic pancreatic ductal adenocarcinoma, paving the way for a Phase 3 trial. The company is also preparing to present additional data from its VIRAGE Phase 2b clinical trial of VCN-01 in metastatic pancreatic cancer at the AACR 2026 Annual Meeting. Theriva Biologics continues to advance its diversified pipeline, positioning itself as a key player in the development of innovative oncology treatments.
